MyoD Directly Up--‐regulates Premyogenic Mesoderm Factors

Description:
Read the paper out of the Tapscott and Skerjanc labs by Gianakopoulos et al., (2011) “MyoD Directly Up--‐regulates Premyogenic Mesoderm Factors during Induction of Skeletal Myogenesis in Stem Cells,” J. Biol Chem 286, 2517--‐2525.
-For this paper, you will need to educate yourself on the following:
 Myogenesis. NCBI Books is a good source for this, particularly the information in Molecular Cell Biology, 4th Ed, by Lodish et al (2004).
 Gain‐of‐function, loss‐of‐function experiments, over‐expression experiments (the text can be helpful for these as can NCBI Books.
 Quantitative PCR – the text has a nice discussion,

1. The hypothesis or question addressed in the experiment(s) of the figure;
2. The punch line conclusion – in most cases this is better than trying to create a mystery narrative unless you have a very specific reason for doing so;
3. The experimental strategy and how it addresses the question or hypothesis;
4. The specific technologies used and how positive results answering the question will be observed (or similarly supporting strongly or negating a proposed hypothesis);
5. A walk‐through of the data and what it shows;
6. The interpretations drawn or conclusions made; and
7. How the interpretations or conclusions lead to the next experiment(s) in the paper.
